Radiation chemistry of nucleotides: 8,5'-cyclonucleotide formation and phosphate release initiated by hydroxyl radical attack on adenosine monophosphates

The formation of 8,5'-cycloadenosine 5'-monophosphate (8,5'/sup -/ cloAMP) in irradiated solutions of adenosine 5'-monophosphate (5'-AMP) has been quantified over the pH range 2 to 10. Maxima for G(8,5'-cycloAMP) occur at pH 3 and 9 in nitrogen- and nitrous oxide-saturated solution. Oxygen completely inhibits the formation of 8,5'-cycloAMP. Over the pH range 3 to 10, G(8,5'-cycloAMP) is approximately equal to G(inorganic phosphate) for 5'-AMP. The pH response of both G(inorganic phosphate) and G(8,5'-cycloAMP) appears to be determined primarily by the reactivity of the various charged forms of the nucleotides to hydroxyl radical attack.
