Chronic effects of inhaled benzene and ingested ethanol on erythroid progenitor cells in C57Bl/6J mice

Exposure of male C57Bl mice for 6 hr/day x 5 day/week to 10 ppm over 178 days or 300 ppm benzene over 60 days in combination with 0% or 5% ethanol in the drinking water 4 days/week for the same time periods produced changes in the in vitro colony forming ability of the erythroid progenitor cells BFUE and CFUE, as well as recognizable erythroid precursors, and mature and red cells. Depressions in marrow CFUE with more severe in mice exposed to 300 ppm benzene in combination with ethanol than in mice exposed to benzene alone. For mice exposed to 300 ppm benzene and ethanol, depressions in marrow and splenic BFUE, CFUE, and normoblasts were followed by transient increases in spleen FLUE, marrow, and spleen CFUE, and normoblasts, and the appearance of nucleated red cells in the peripheral blood. In contrast, only marrow normoblasts and spleen CFUE increased following initial depressions in mice treated with 300 ppm benzene alone.
