Effect of dosage and form of cadmium on its absorption and distribution

The retention of and tissue distribution of Cd administered as CdCl/sub 2/ or Cd-thionein (Cd-TH) was determined. The tissue distribution of Cd was determined 7 days after oral administration of various dosages of CdCl/sub 2/ (1-10,000 ..mu..g Cd/kg) to rats. The results indicated that the concentration of Cd in tissues increased more than the increase in dosage. To determine the gastrointestinal absorption of Cd, rats were given an oral dosage of Cd and 3 hours later organs were removed to determine Cd content. Concentrations of Cd in tissues again increased more than the increase in dosage. These results suggest that retention of Cd following oral administration is dosage-dependent and results from increased absorption of Cd at higher dosages. However, the binding of Cd to metallothionein does not appear to influence the dosage-dependent absorption of Cd. The distribution of Cd as CdCl/sub 2/ was also compared with the tissue distribution of orally-administered Cd-TH. Tissue concentrations of Cd indicated that less Cd was absorbed as Cd-TH than CdCl/sub 2/. Although administration of Cd-TH resulted in higher kidney/liver Cd concentration ratios than CdCl/sub 2/ (9 vs 2), renal Cd concentrations were similar to those obtained after CdCl/sub 2/ administration. These results indicate that the high kidney/liver Cd concentration ratio after Cd-TH treatment versus CdCl/sub 2/ is due to lower concentrations of Cd in liver rather than marked increases in renal Cd levels. In conclusion, both the dosage and chemical form of Cd are important factors in determining the absorption and distribution of Cd. 11 references, 6 figures.
