ESR of /sup 125/IUdR-labeled cells and x-irradiated cells at 4. 2/sup 0/K and above

The high toxicity of /sup 125/I-deoxyuridine (/sup 125/IUdR) to cells was studied by comparing the electron spin resonance (ESR) spectra of cells labeled with /sup 125/IUdR and cells that have been irradiated with x rays. The kinds of radicals produced by the two types of radiation appear to be the same, but there are differences in relative amounts in three cases: (1) several times as many carbon-based radicals relative to the amount of radiation products of cellular water are produced in the /sup 125/I-labeled cells as in the x-irradiated cells. This indicates that the indirect effect due to the radiation products of water are probably of less importance in radiation cell lethality of /sup 125/I-labeled cells than the direct effects; (2) there is more of a 17-G doublet present in the ESR spectra of /sup 125/IUdR-labeled cells than in the x-irradiated cells. This doublet is probably due to the thymine anion radical; and (3) there is more of a radical, which is probably the thymine H-addition radical, present in the /sup 125/IUdR-labeled cells. These results strengthen the hypothesis that the high toxicity of /sup 125/IUdR is due to a high local ionization density at the DNA.
