This patent describes an assay method for detecting a nucleic acid analyte in a sample, employing two sets of reagents, a labelling set and a capturing set. The method comprising: (1) providing members of the labelling set of reagents; providing members of the capturing set of reagents, combining in a liquid medium under binding conditions for complementary pairs, the sample containing analyte in signal-stranded form, with the labelling and capturing probes, to provide probe-analyte complexes; combining the probe-analyte complexes with the capturing reagent to give support-bound probe-analyte complexes; washing the support to provide the support-bound probe-analyte complexes substantially free of unbound probe and unbound analyte; contacting the support with the labelling reagent to label the support-bound probe-analyte complexes; separating the label into a bound phase and an unbound phase by means of the solid support; and detecting the amount of bound or unbound label as determinative of the presence of analyte.
