Nucleic acid hybridization assays

Patent ·
OSTI ID:6719852

This patent describes a method for carrying out a nucleic acid hybridization test to detect a target nucleic acid sequence. It comprises: providing a polymer molecule bonded to a first single stranded nucleic acid sequence comprising a first probe which hybridizes to a first target nucleic acid sequence when present, provided that, where the polymer molecule comprises DNA, the DNA is of heterogeneous base sequence; denaturing the target nucleic acid sequence; contacting the denatured target nucleic acid sequence with the polymer molecule bonded to the first single stranded nucleic acid sequence; separating out complexes between the target nucleic acid sequence and the polymer molecule by trapping the complexes on a two-dimensional surface; and detecting complexes between the target nucleic acid sequence and the polymer molecule.
