GRAFT-VERSUS-GRAFT IMMUNOLOGICAL TOLERANCE IN RADIATION CHIMAERAS

In a system of two different hybrid grafts of restorative embryonic tissue given to lethally irradiated CBA mice the immune status of the established chimaeras was studied, especially regarding the concept of graft-to-graft tolerance. The results of skin grafting and tests for spleen and liver enlargement induced with chimaeric spleen cells are in agreement with the idea that under certain conditions the embryonic graft can acquire tolerance to foreign antigens met in the host environment. The rate of immunological maturation of the embryonic graft in the environment of the irradiated adult host was studied in the graft-to-graft system using a direct cytological follow-up of the fate of chromosome marked cells. The experimental results are interpreted as evidence of slower immunological maturing and therefore of a prolongation of the tolerance-responsive phase of the embryonic graft under the conditions of the experiment. The possible role of some factors of a nonimmune nature which may influence the relations, between restorative cells of different origin in radiation chimeras is discussed. (auth)
