ADOPTIVE TOLERANCE: TRANSFER OF THE TOLERANT STATE

Tolerance to skin homografts in C3H mice was induced by intravenous neonatal injection of CBA spleen cells, and the lymphoid cells from the tolerant mice were transferred to lethally (750-r) x-irradiated recipient mice of both host and donor strain. The irradiated recipient mice were subsequently tested with appropriate skin grafts to determine whether the tissues transferred from the tolerant animal continue their tolerant behavior in a new environment. The fate of skin grafts from a third unrelated strain (C57BL6) of mice on the chimeras determined whether the transferred cells, although mutually tolerant, were still capable of an immunologic response. The results showed that lymphoid cells from tolerant mice can be successfully transferred to a new environment without affecting their tolerant behavior. In addition to being mutually tolerant, the host and donor cells from the tolerant animal tolerated each other's type of skin grafts. The tolerance was not due to a general impairment of the immune capacity but to a specific inhibition of the immunologic response. The results suggest that the graft-vs-host tolerance in the newborn C3H mouse injected with adult CBA strain spleen cells was not due to immunologic paralysis. (TCO)
