KINETICS OF RETICULOENDOTHELIAL PHAGOCYTIC RESPONSE TO INTRAVENOUSLY ADMINISTERED Au$sup 198$ COLLOIDAL GOLD IN RABBITS

Phagocytosis of colloidal radiogold-Au/sup 198/, observed in the intact animal by external scintillation detection techniques, proceeds according to kinetic relations which are compatible with direct observations of the physiological process. The cellular engulfment of colloidal-sized material by the von Kupffer cells of the liver was described to occur in two stages; adherence of the particle to the surface of the cell, and then engulfment. Rate measurements of the blood disappearance and of the hepatic deposition of colloidal radiogold-Aui/sup 198/ are shown to conform with the kinetic model of the Michaelis-Menten relation; a reversible reaction complex is formed by combination of a reactant with a reaction site (such as a reaction center on an enzyme) and a nonreversible reaction follows. This parallelism between description of an event and the kinetics determined indirectly recommends reliability of the experimental method employed here for investigation of the phagocytic capacity of the von Kupffer cells of the reticuloendothelial system. These experimental measurements also conform with concepts developed for disappearance of I/sup 131/-tagged denatured serum albumin and carbon particles from the blood stream reported by Halpern and coworkers.
