A study was made of whether bilirubin is oxidized by the free radicals forming during the radiolysis of water containing this pigment, and, if so, whether this can be prevented by radioprotective substances. Bilirubin solutions of various concentrations of pH 5.8, 7.35, or 13,3 were irradiated at a rate of 236 r/sec, In solutions containing 10 mg/l the pigment was destroyed at doses above 5000 r and this was 98% complete at 100,000 r. Similar results were obtained at all 3 pH values. The following substances protected bilirubin, some of them at concentrations as low as 0.001M; ascorbic acid, cysteine, cystine, e rgothionine, 5-hydroxytryptamine, propylthiouracil, methylthiouracil, thiouracil, tryptamine, cysteamine, cystamine, 5-hydroxyindoleacetic acid, thiosalicylic acid, and thioacetamide. Thiourea, urea, and glucose were ineffecttve at 1000- fold higher concentrations. Most of the compounds affording radioprotection to bilirubin contain labile H atoms. Radio-oxidation of btlirubin does not give rise to btliverdin, but probably to a product similar to that formed by direct oxygenation, (H.H.D.)
