Protein-free solutions of hemin were irradiated with soft x rays (50 kv) at a dose rate of 2065 r/sec. Various radioprotective agents were added to the solutions, which were buffered at pH 7.35 or 13.33, and the percentage of hemin recoverable after irradiation was determined. Destruction of hemin was detected at doses above 50 kr and was 80 to 85% complete at 0.5 to 1 Mr. Destruction was greater at pH 7.35 than at 13.33. In the presence of radioprotective agents at concentrations of 3 x 10/sup -9/ M the percentage destruction of hemin at 0.5 Mr was as follows: ascorbic acid 30%, cystine 60%, cysteine 70%, cystamine 75%, and cysteamine 80%, compared with 85% in their absence. The presence of plasma, bovine serum albumin, or globin almost completely prevented breakdown of hemin during irradiation. H/sub 2/S formed during the irradiation of cysteine was shown to react with hemin. It is suggested that radiation, as do oxidizing agents, split hemin into 2 fragments. (H.H.D.)
