DISAPPEARANCE OF TRITIATED THYMIDINE AND TRITIATED WATER FROM THE ANTERIOR CHAMBER OF THE RABBIT EYE

The rate of disappearance of injected tritium-labeled thymidine from the anterior chamber of the rabbit eye was measured. Parallel experiments were performed to determine the rate of disappearance of tritiated water under similar conditions. Young American Dutch rabbits were used in all experiments. After phenobarbital anesthesia 0.1 ml of aqueous humor was withdrawn from the anterior chamber and replaced with 0.1 ml of tritiated thymidine at a concentration of 1 mu c/ml in Eagle's basal medium with a specific activity of 1.90 c/mm or tritiated water at 40 mu c/ml. Samples were withdrawn at intervals of 2.5 to 120 min and counted in a scintillation spectrometer. Results show a rapid disappearance of tritiated thymidine and of tritiated water from the anterior chamber. It is concluded that the availability of tritiated thymidine in the aqueous humor, following injection irto the arterior chamber, is of short duration. The use of tritiated thymidine as a tracer in studies on the synthesis of desoxyribonucleic acid in the eye is discussed. (C.H.)
