OXIDATIVE PHOSPHORYLATION IN THE LIVER AND KIDNEY OF RATS TREATED WITH FRACTIONS OF HOMOLOGOUS HOMOGENATES OF ORGANS OF IRRADIATED ANIMALS

When whole homogenates of various organs removed from rats exposed to 400- or 800-r doses of whole-body x irradiation were injected intraperitoneally in normal rats, liver and kidney mitochondria isolated from the recipient rats showed an inhibition of oxidative phosphorylation, or a diminished P/O ratio. The 400- and 800-r doses corresponded to the LD/sub 50/ and LD/sub 100/ doses in the animals. In attempts to isolate the inhibitory factor in homogenates of irradiated organs, the homogenates were fractionated by high-speed centrifugation in a sucrose density gradient into nuclear, mitochondrial, and supernatant fractions. The fractions were then injected intraperitoneally in normal rats, from which liver and kidney mitochondria were isolated and assayed for oxygen and inorganic phosphate uptake. Almost all of the inhibitory action of irradiated homogenized organs could be found in the supernatant fraction; nuclei and mitochondria showed little activity. For example, the supernatant from 250 mg (fresh weight) liver or kidney irradiated with either x ray dose reduced the P/O ratio of liver mitochondria of the recipient rats to values of 1.83 to 1.90 compared with a control value of 2.81. In kidney mitochondria the corresponding values were lowered to 1.76 to 2.06 compared with 2.89. Whole homogenates and the supernatant fraction induced swelling of liver and kidney mitochondria but this was not seen with injections of nuclear and mitochondrial fractions. (H.H.D.)
