EFFECTS OF X IRRADIATION ON THE MUCOPOLYSACCHARIDES IN THE AORTIC WALL

>The effects of irradiation on aortic mucopolysaccharides were investigated in vitro and in vivo. Injection pressure in media and intima drops instantaneously during in vitro irradiation of aorta with 1000 r, indicating an increased water permeability. Diffusion of dyes into the intimal layer occurred at an increased rate in x irradiated segments of excised aorta (human, cow, pig, dog). Injection of dye solutions in rats after whole-body irradiation (3000 r), followed by spectrophotometric estimation of dye content in the intimal membrane, demonstrated an increased accumulation of dye (trypan blue, Evans blue, hematoporphyrin) in comparison with measurements on aortas of nonirradiated control animals. In experiments with rabbits fed on a high cholesterol diet, irradiation with 100 r weekly for 10 weeks resulted in a higher incidence of atheromatous lesions, compared with nonirradiated animals fed the same diet. The presence of Na/sub 2/S/sub 2/O/sub 3/ provided an effective protection against the drop of injection pressure and the increased diffusion rate of dyes after irradiation. It is suggested that the described vascular radiation efiects may contribute to the shortening of lifespan after subletual and chronic irradiation and that Na/sub 2/S/sub 2/O/sub 3/ may have protective value against these effects. The results suggest that mucopolysaccharide depolymerization is caused by x irradiation, which is reflected by increased permeability of the aorta. (TCO)
