The Action of Chlordiazepoxide on Metabolism and Body Temperature of the Mouse: Data on the Mechanism of Action of Radioprotective Agents
This drug, Librium or 7-chloro-2-methylamino-5-phenyl1,4-benzodiazepine- 4-oxide-hydrochloride, when given to mice in doses of 7.5 to 50 mg/kg had a biphasic effect on oxygen consumption. Consumption fell sharply 15 min after its injection, then rose at 30 min above the basal level. Injection of the drug at high concentrations inhibited respiration of brain, liver, and diaphragm, but when given in vivo at a dose of 25 mg/kg, respiration in these tissues was not affected. Injection of mice with doses above 7.5 mg/kg reduced body temperature for a 2-hr period; at the highest dose (50 mg/kg) the temperature fell 5 deg C. This drug when given to irradiated mice reduces the mortality rate from 100% to 25 to 50%, and in this respect it resembles other drugs which produce hypothermia and slow respiration. This was demonstrated when the mice were injected with 8.75 mg/kg 15 min before irradiation with 800 r at a dose rate of 40 r/min. The radioprotective effects of the compound are attributed to both its ability to lower body temperature and diminish respiration rate. (H.H.D.)
