PROTECTIVE ACTION OF ETHYL PALMITATE AGAINST ROENTGEN RADIATION (in German)

Earlier studies showed that splenectomized animals recover faster from radiation injury than controls with intact spleens, and that intravenous injection of ethyl palmitate results in destruction of the spleen. It might be expected that chemical splenectomy, which is less stressful to the animal than the surgical operation, might confer significant radioprotection. In preliminary toxicity studies with mice, it was found that the degree of purity of ethyl palmitate, injected intravenously as an emulsion, is of great importance. Injection of 100 mg of the technical grade resulted in death of 20% of the animals within 12 hr, whereas 80% died after injection of a similar amount of the purified fat. Female mice were more sensitive to the fat than males. Animals surviving for 24 hr after injection appeared normal and were used for radiation experiments. A considerable radioprotective effect was obtained by intravenous injection of ethyl palmitate 48 hr before 690-r irradiation; 53% more injected mice survived than uninjected controls. The degree of purity of the preparation used did not influence its radioprotectivity. Further experiments showed that the protective action increased with injected dose over the range 25 to 100 mg. Surgical splenectomy 48 hr before irradiation was found to be almost as effective as injections of the fat. The mechanism of the protective effect of the fat was not clear since maximum effectiveness was reached 48 hr after the injections. Further studies will be required to determine whether its effect is a nonspecific one rather than one manifested through the reticuloendothelial system or the adrenal cortex. (BBB)
