ACTION OF CYTOTOXIC SERUMS ON WHOLE-BODY Co$sup 60$-IRRADIATED ANIMALS. II. HISTOLOGIC FINDINGS (in Italian)

In the first report of this series it was shown that the lethal effect of whole-body Co/sup 60/ gamma irradiation of guinea pigs was accentuated by subsequent injection of cytotoxic antiserum. The antiserum was prepared by intravenous injection of rabbits with a suspension of guinea pig lymphoid cells, which gave titers up to 1: 128. Histologic changes in the lymph nodes are reported for these guinea pigs, which were irradiated with 200, 400, or 600 r and injected intraperitoneally with 0.25 ml of this antiserum every 3rd day postirradiation. About 30% of unirradiated animals so injected died within 30 days, presumably on account of anaphylaxis, showing congestion and eosinophil infiltration of spleen and other lymphatic tissues. Injection of cytotoxic serum after 200 r resulted in 100% mortality by 21 days whereas normal rabbit serum after 200 r caused only 20% 30-day mortality. The respective rates after 400 r were 100% at 15 days and 60% at 30 days, and after 600 r, 100% at 8 days and 100% at 30 days. In animals injected with an antiserum and irradiated with 400 or 600 r, complete destruction of the spleen was evident by the 6th to 8th day, which was followed by fibrotic replacement. These changes were much more severe and rapid than after irradiation alone. The results indicated that panirradiation at doses of 200 r and above markedly inhibits the defense mechanisms of the guinea pig against cytotoxic serum, allowing the anticellular antibodies contained therein to augment the injurious action of radiation on lymphoid cells.
