The right eye of adult male rats of the Osborne-Mendel strain were irradiated with a single 1000-r dose of x rays; both eyes were removed 1 hr to 28 days later and examined histologically. At 1 hr postirradiation the postequatorial zone of the lens showed no changes, but in the preequatorial and equatorial zones the number of mitoses was 10% less than that in the left lens. Atypical anaphase and telophase figures and unequal distribution of chromosomes were seen in the right lens epithelium. At 2 hr, degenerative and lytic changes were seen in nuclei in the postequatorial zone, and in the other zones, mitoses were further diminished in number. By the 4th hr, nuclear fragmentation was evident, and the 1st changes were present in the central zone of lens epithelium. Degenerative changes in nuclei continued to dominate the histologic picture for the 1st week postirradiation, following which there were acellular areas in the epithelium. After the 2nd week, numerous atypical mitoses were seen, giving rise to giant nuclei. These nuclei were 2--3 times normal size, were only slightly granulated, and stained weakly by the Feulgen method. By the 4th week the pre- equatorial and equatorial zones showed large, confluent acellular regions, with giant nuclei scattered throughout the cellular regions. The central zone of the epithelium appeared to be the most radioresistant and the equatorial zone the most radiosensitive.
