ON THE EFFECTS OF FRESH AND DEEP-FROZEN PRESERVED ISOLOGOUS BONE MARROW ON THE HEMATOLOGIC RADIATION SYNDROME AFTER LETHAL WHOLE-BODY IRRADIATION OF RATS (in German)

Thirty-five white rats (Sprague-Dawley) were total-body irradiated with 1000 r Co/sup 60/ gamma rays. The effect of an isologous bone marrow cell transfusion (2.5 x 10/sup 6/ cells/100 g, given intravenously 24 hr after the irradiation) upon the hematologic radiation response was investigated. Except for one control group, which received no marrow cell transfusion, the animals received either freshly taken or preserved (at -70 deg C in dimethyl sulfoxide) bone marrow. With this, the fatal results of the total-body irradiation could be inhibited consistently. Body weight showed a continuous increase 5 days after the radiation application in the animals treated with bone marrow. The control group developed rapidly anemia, leukopenia, and thrombopenia which remained stationary until death. After bone marrow transfusion the blood cell values were normalized within about 4 weeks. Leukopenia lasting 2 to 3 weeks could not be inhibited even by administration of marrow cells. A transient anemia was found in the animals which were treated with fresh bone marrow but not after injection of the cells preserved in dimethyl sulfoxide. The primary thrombocyte decrease could be inhihited by administration of native cell suspension. Using preserved material the thrombocytes had apparently lost their viability but thrombopoiesis by the megakaryocytes was probably still present. (auth)
