Radioimmunoscorbent assay of follicle-stimulating hormone in serum and urine from men and women

A radioimmunosorbent technique was used for the assay of the pituitary gonadotropins. Human follicle-stimulating hormone (FSH) and luteinizing hormone (LH) preparations of the highest purity and labeled with /sup 125/I were used as reference standands. The antisera were raised against more purified gonadotropin preparations than generally used as immunogens. The immunosorbent preparations with antibodies directly or indirectly coupled to insoluble polysaccharides were tested for their capacity to bind labeled antigen during 24 hrs in a total reagent volume of 0.7 ml. The amount that bound about 15% was determined and used in the assays. The use of solid phase-coupled antibodies simplifies the separation of antibody-bound and free antigen. The separation can be made highly effective with an almost complete removal of unbound antigen. It is possible to obtain a rapid answer within 27 hrs without sacrificing high sensitivity and precision. The method is well suited for large scale analyses of protein hormones in the clinical routine. Physiological studies on episodic changes of FSH and LH in serum in the daytime, on FSH and LH in serum and urine during the menstrual cycle, and on serum levels and urinary excretion of FSH and LH were performed.
