Double antibody solid phase radioimmunossay for human FSH (in Japanese)

Double antibody solid phase (Dasp) was applied to the radioimmunoassay for follicle-stimulating hormone (FSH) in the human seram. Materials consisted of /sup 131/I-labeled purified human pituitary FSH (LER 1366), LER 907 which was the pituitary standard of FSH and lutenizing hormone (LH), 2nd 1RP-HMG, and antihuman pituitary FSH serum (Anti-HPFSH). Human chorionic gonadotropin (HCG) (12,000 IU/mg) and human pituitary LH (LER 960) were employed to examine the specificity. After the first incubation, 1 ml of anti-RGG immunosorbent (1: 20) was added to the tube and it was further incubated (2nd incubation). The first incubation was continued for 96 hrs at 4 deg C and the second was for 2 hrs at room temperature. Reaction curves of various hormones to the dosage in these measurements were obtained, and the curve of LER 1366, LER 907, IRP-HMG, and the serum of women were parallel to each other. LER 960 did not react by the administration of less than 60 mg/ml and the purified HCG of Donini also did not react. Therefore, these measurements were not affected by LH and HCG, which were physiologically present in the blood. The serum FSH of a 24 year-old patient with opsomenorrhea, who was pregnant by the administration of clomiphene, was continuously determined during two cycles of these measurement methods. Dasp method was revealed simple to separate bound and free hormones. (JA)
