Evaluation of section thickness of skin in determining the epidermal labeling index with tritiated thymidine

Epidermal basal cells in the albino guinea pig were pulse labeled with tritiated thymidine (/sup 3/H-TdR) by intraperitoneal or intradermal injection, and the fraction of labeled cells STAlabeling index (LI)! was compared in 4 mu versus 6 mu sections. The LI scored in 6 mu sections was about 10% lower than that in 4 mu sections. For our purpose, it is unnecessary to submit to the technical difficulties of routinely cutting sections of skin thinner than 5 to 6 mu for autoradiography of /sup 3/H-TdR, since the increase in scoring efficiency is relatively small (10 to 20%).
