Effect of certain hormones on the inclusion of $sup 32$P in the phospholipids of the intestinal mucosa. I. The role of insulin (in Bulgarian)
An experiment was carried out with 20 broilers age 2 to 2.5 months. Seven of them were used as controls; 6 were treated in the course of 3 days consecutively with insulin at a rate of 3 ME/kg live weight; the remaining 7 chickens were treated with 250 mg alloxan/kg live weight. In the bilaterally isolated jeunum, with 20 cm length of the segment, were introduced 20 millicurie/ kg live weight of KM/sup 32/PO/sub 4/. Two hours later the isolated intestinal segment was removed and the lipids were extracted from the mucosa by means of chloroform-methanol (2: 1). The separate phospholipid fractions were divided by thin layer chromatography, on silica gel G and after developing them by means of iodine vapors the spots were scraped in separate pots. The radioactivity was measured with the aid of a Geiger-Mueller counter. The total phospholipids were counted from the lipid extract from which two samples were prepared of 0.3 ml each. For higher accuracy two parallel chromatograms were prepared. The blood sugar level was also determined after treatment with insulin and alloxan. The data obtained show that: the insulin certainly (P < 0.001) raises the level of the total phospholipids in the jeunum mucosa of the chickens. A certain trend was observed of reducing the inclusion of /sup 32/P in the total phospholipids under the effect of alloxan. A reverse correlation was established between the sugar content in the blood and the level of the total phospholipids. The highest rate of inclusion of /sup 32/P under the effect of insulin was observed in the lecithin (P < 0.001), followed by sphingomyelin (P < 0.002), lysolecithin (P < 0.10) and cephalin (P < 0.002). The cardiolipin is without essential changes. The reduction of the phospholipids under the effect of alloxan is mainly on account of the lysolecithin while cardiolipin remains unchanged. (auth)
