PREPARATION OF BILIRUBIN LABELED WITH I$sup 13$$sup 1$

Iodination of bilirubin was carried out using I/sup 131/. Both iodinated bilirubin and biliverdin were obtained, yields ranged from 7-- 16 percent and 10-- 25 percent, respectively. The purity of each radiocompound was determined chromatographically. Normal human serum albumin labeled with radioiodinated bilirubin was prepared and lyophilized. The bilirubin-albumin complex was assayed 24, 48, and 72 hours after preparation. The final product of bilirubin iodination by this technique is a mixture of bilirubin derivatives, one of which is biliverdin. Separation provides a labeled bilirubin that resembles uniodinated bilirubin in physical, chemical, and physiologic characteristics. The method is a relatively inexpensive way of preparing bile pigments that may be used in bilirubin metabolism studies and in liver function tests. (H.M.G.)
