Previous research on the survival of yeast cells showed that haploid yeast cells are the most sensitive to radiation, and diploid yeasts are the most resistant to radiation. In most cases, a microorganism that cannot grow on a medium lacking some essential ingredient carries a point mutation, and can revert to its initial wild state. It would be expected that with the same irradiation dose, the mutation reversal would have twice the frequency in the homozygous diploid mutant than in the haploid mutant. To test this hypothesis, haploid and diploid yeast cells were irradiated with Co/sup 60/ gamma rays and ultraviolet radiation. The results showed that the number of mutation reversals in the haploid stralns increased more rapidly than in the corresponding isogenous diploid strains with increasing radiation dose. The behavior was similar for the two types of radiation. The increased frequency of mutation reversals of haploid cells as compared to diploid cells with the same irradiation dose can be partially explained, if it is assumed that a number of sites in the gene have been modified and can act as suppressors. This type of suppressor is recessive. (TTT)
