THE IN VIVO STABILITY OF THE TRITIUM LABEL IN 1,2-H$sup 3$-D-ALDOSTERONE WHEN USED FOR MEASUREMENT OF ALDOSTERONE SECRETION RATE BY THE DOUBLE ISOTOPE DILUTION TECHNIQUE
A mixture of D-aldosterone and 1,2-/sup 3/H-D-aldosterone was prepared, and part of this mixture was injected intravenously into two totally adrenalectomized patients. The remainder was retained, its specific activity was determined, and compared with the specific activity of the 3-oxoconjugate of aldosterone which was recovered from urine. The specific activity of the aldosterone released by pH 1 incubation of the urine should be lower than that of the uninjected aldosterone if some of the tritium tag is removed during in vivo metabolism, or if the patients had some residual adrenal tissue capable of producing endogenous aldosterone. The tritium label of 1,2-/sup 3/H-D- aldosterone is biologically stable and this compound can be used satisfactorily for the determination of aldosterone secretion rates in man. (BBB)
