Mechanisms of dissolution of nickel subsulfide in rat serum

To elucidate the initial steps of carcinogenesis by nickel subsulfide, the mechanisms of dissolution of ..cap alpha..-Ni/sub 3/S/sub 2/ were studied by incubating ..cap alpha..-Ni/sub 3/S/sub 2/ dust in (a) water, (b) rat serum, and (c) rat serum ultrafiltrate. The sediments that remained following incubationn of ..cap alpha..-Ni/sub 3/S/sub 2/ in the 3 media for 2 weeks were examined by x-ray diffractometry. The rates of solubilization of /sup 63/Ni from ..cap alpha..-/sup 63/Ni/sub 3/S/sub 2/ during incubation in the 3 media were measured by liquid scintillation counting. These studies showed that solubilization of Ni(II) from ..cap alpha..-Ni/sub 3/S/sub 2/ in rat serum requires the presence of O/sub 2/ and involves three reactions: (I) 2..cap alpha..-Ni/sub 3/S/sub 2/ + O/sup 2/ + 2H/sub 2/O ..-->.. 4 ..beta..-NiS + 2Ni (OH)/sub 2/; (II) ..beta..-NiS + 2O/sub 2/ ..-->.. Ni/sup 2 +/ + SO/sub 4//sup 2 -/; and (III) Ni/sup 2 +/ and Ni(OH)/sub 2/ + serum ligands (albumin and amino acids) ..-->.. soluble Ni(II)-complexes. The rate of solubilization of /sup 63/Ni from ..cap alpha..-/sup 63/Ni/sub 3/S/sub 2/ was initially more rapid in rat serum than in serum ultrafiltrate and much more rapid than in water. After 2 days of incubation, the rates of dissolution of ..cap alpha..-/sup 63/Ni/sub 3/S/sub 2/ in the 3 media became progressively retarded, probably owing to deposition of insoluble surface coatings of ..beta..-/sup 63/NiS and /sup 63/Ni(OH)/sub 2/ on the ..cap alpha..-/sup 63/Ni/sub 3/S/sub 2/ particles.
