Radioligand assay for angiotensin II receptors

An assay procedure for measurement of angiotensin II (AII) receptors with monoiodinated /sup 125/I-angiotensin II in partially purified adrenal membranes and isolated adrenal cells is described. The same general principles can be applied to the assay of receptors with (/sup 3/H)angiotensin II and radiolabeled agonist and antagonist analogs of the octapeptide. The Asp or Asn, and Val or Ile forms of AII, can be used for radioiodination to prepare tracer suitable for receptor binding studies. For some purposes, the Sar derivative of AII is preferable for labeling, since it has higher affinity for AII receptors and may provide higher specific binding of tracer when small numbers of receptor sites are to be analyzed.
