Effect of dietary copper on platelet volume and cytochrome c oxidase (CCO) in platelets

Results from a recent study with young women indicated that CCO activity in PLT may be a sensitive indicator of Cu status. Furthermore, abnormal PLT and CuD have been associated with atherosclerosis. Thus, to examine the effect of CuD on PLT variables, weanling male Sprague Dawley or Fisher rats were fed Cu deficient (CuD <1 ppm) or adequate (6 ppm Cu) diets. After 5 weeks, several indices of Cu status, including liver and plasma Cu, and ceruloplasmin were lower in CuD. PLT Cu, CCO and superoxide dismutase (SOD) were lower, and mean platelet volume (MPV) was increased in CuD. In a separate study, CCO activity in PLT was directly correlated with Cu intakes below 5 ppm. These data indicate that CCO in PLT is a sensitive indicator of Cu status and that energy metabolism and resistance to oxidative damage are impaired in PLT from CuD animals. The apparent increased MPV seen in CuD and atherosclerosis may be the result of fewer small PLT. Larger MPV, seen in CuD and atherosclerosis, may lead to increased aggregability and clot formation.
