Assignment of the human carboxypeptidase E (CPE) gene to chromosome 4

We recently reported the characterization of a human and a rat brain cDNA-encoding carboxypeptidase E (CPE or carboxypeptidase H; EC 3.4.17.10) isolated during our screening for brain expressed genes. The human and rat CPEs share more than 95% protein sequence homology with bovine CPE. CPE is a processing enzyme that cleaves C-terminal amino acids and is implicated in biogenesis of neuropeptides and neurohormones, being enriched in brain and adrenal secretory granules. Human CPE is highly related to human carobxypeptidases N (51% identity) and M (43% identity), which occur in serum and ubiquitously in cell membranes, respectively. The functions of these other carboxypeptidases is not yet known. The less related carboxypeptidase A has been mapped to chromosome 7q (320ter) as has trypsin. We now report the localization of CPE to chromosome 4, to which other proteases also map as well as several genes of neurological interest.
