High performance capillary electrophoresis-electrospray ionization mass spectrometry of nucleotides

In this presentation, the on-line coupling of high-performance capillary electrophoresis to electrospray ionization mass spectrometry (ESI-MS) is reported. In the beginning stage, capillary zone electrophoresis (CZE)-mass spectrometry was used to separate a group of monophosphate nucleosides. The radiation-damaged products only count about 0.3% of original sample. CZE-MS could not provide enough resolution and sensitivity to distinguish the damaged and native nucleotides with MS-compatible buffer systems although the encouranging separation of model solutes was obtained. By utilizing the zone self-sharping and concentrating effects of capillary isotachophoresis (CITP), the promising separation and identification of native and damaged nucleotides and free bases using CITP-MS was realized.
