Radioimmunoassay for leukotriene D/sub 4/ leukotriene E/sub 4/
A conjugate of leukotriene (LT) D/sub 4/ and keyhole limpet hemocyanin was made using difluorodinitrobenzene as link to the free amine on LTD/sub 4/. This conjugate was emulsified in Freund's adjuvant and injected intradermally into New Zealand white rabbits at two week intervals. Using charcoal to separate bound from free and /sup 3/H-LTD/sub 4/ as the ligand, the antiserum obtained after 6 months displays 100% cross-reactivity between LTD/sub 4/ and LTE/sub 4/, 5.3% with LTC/sub 4/ and negligible affinities for other arachidonic acid metabolites or glutathione. The /sup 3/H-LTD/sub 4/ is displaced by 50% with 75pg of LTD/sub 4/ or LTE/sub 4/ and the detection limit of this assay is 12.5pg. Since LTE/sub 4/ is the major leukotriene metabolite present in human urine, this RIA may be useful in determining the daily excretion of leukotrienes.
