Free radical formation during in vivo metabolism of Freon 11 (CCl/sub 3/F) and 1,1,1-trichloroethane

Free radical products were identified by electron paramagnetic resonance (EPR) techniques in rat liver lipid extracts 2 hours after administering the refrigerant Freon 11, CCl/sub 3/F (3-7% in air) or the industrial solvent 1,1,1-trichloroethane (0.5%) by inhalation. Phenyl-t-butyl nitrone (PBN) was used as the spin-trapping agent and was given in a corn oil-phosphate buffer emulsion by stomach tube prior to exposure to CCl/sub 3/F or 1,1,1-trichloroethane. The EPR spectrum of the PBN-trapped radical derived from CCl/sub 3/F metabolism, consisted of a triplet of triplets. This indicates the presence of a fluorine atom attached to the trapped carbon-centered radical. Splitting constants for the PBN-trapped radical derived from 1,1,1-trichloroethane metabolism were A/sub N/=14.81 G and A/sup H/=2.08 G. When metyrapone, an inhibitor of the 52 kd liver cyt. P-450, was administered i.p. one hour before the PBN and exposure to the CCl/sub 3/F or 1,1,1-trichloroethane, the formation of PBN-trapped radicals was inhibited 80%.
