Title: Synthesis of a precursor for the preparation of 9 alpha,11 alpha-tritiated 5 alpha-androstane-3 alpha,17 beta-diol 17-glucuronide

Abstract

Starting from 11 beta-hydroxytestosterone, the synthesis of a strategic precursor, C-9 (11) unsaturated 5 alpha-androstane-3 alpha, 17 beta-diol 17-glucuronide (9a), for the preparation of 9 alpha,11 alpha-tritiated 5 alpha-androstane-3 alpha, 17 beta-diol 17-glucuronide has been achieved. The authors optimized the reaction conditions for catalytic reduction employing hydrogen and subsequent base hydrolysis followed by purification on Amberlite XAD-2 resin to obtain the saturated 5 alpha-androstane-3 alpha, 17 beta-diol 17-glucuronide.

  • A simultaneous radioimmunoassay procedure for plasma testosterone (T), 5alpha-androstan-17beta-ol-3-one (DHT), 5alpha- androstane-3alpha,17beta-diol (3alphaol), and 5alpha-androstan- 3beta,17beta-diol (3betaol) is described in this report. Peripheral plasma concentrations of T, DHT, 3alphaol, and 3$beta$ol were 1.61 +- 0.26, 0.49 +- 0.15, 0.17 +- 0.03, and 0.24 +- 0.04 ng/ml, respectively, in adult male rabbits. In contrast, T, DHT, 3$alpha$ol, and 3$beta$ol peripheral plasma concentrations in dexamethasone-treated castrate rabbits were 0.07 +- 0.001, 0.02 +- 0.01, 0.07 +- 0.01, and 0.05 +- 0.01 ng/ml, respectively. These results represent the first simultaneous measurement of T, DHT, 3$alpha$ol, and 3$beta$ol in peripheral plasma of any malemore » vertebrate.   • Sertoli cells from immature rats metabolized (/sup 3/H) 5 alpha-androstane-3 alpha, 17 beta-diol to (/sup 3/H) 5 alpha-androstane-3 alpha, 16 alpha, 17 beta-triol and (/sup 3/H) 3 alpha-hydroxy-5 alpha-androstan-17-one. This is the first report of 16 alpha-hydroxylation of 5 alpha-reduced androgens in the testis. FSH significantly stimulated 16 alpha-hydroxylation while LH significantly decreased this activity. 3 alpha-Hydroxy-5 alpha-androstan-17-one was the major metabolite formed and its production was significantly increased in the presence of both LH and FSH, although FSH stimulation was significantly more than LH.   • A rapid specific and reliable RIA for urinary 5..cap alpha..-androstane-3..cap alpha.., 17..beta..-diol (Adiol) is described using chromatographical purification and a specific antibody. Values are reported under some physiological and pathological conditions in 179 individuals. In 43 normal adult men the mean (+- SD) urinary Adiol excretion was 193 +- 77 ..mu..g/24 h, and in 29 normal women it was 44 +- 23 ..mu..g/24 h. These values are significantly different (P < 0.01). In 49 hirsute women, urinary Adiol Excretion was elevated (137 +- 51 ..mu..g/24 h) and significantly different from this value in normal women (P < 0.01). The urinarymore » Adiol excretion in 10 postmenopausal women was very low (< 5 ..mu..g/24 h). In normal adult subjects, the theoretical contribution to urinary Adiol of the major secreted androgens was calculated. Whereas dehydroisoandrosterone and dehydroisoandrosterone sulfate yield the same amount of urinary Adiol in both sexes, testosterone is the main precursor of Adiol in men and androstenedione is the main precursor in normal premenopausal and hirsute women. However, the amount of Adiol recovered in the 24-h urine depends not only on the secretion rate of androstenedione and testosterone but is also related to the testosterone 5..cap alpha..-reductase activity present in androgen target cells, especially in sexual skin.« less
  • Two 17 alpha-(/sup 125/I)iodovinyl estradiol derivatives 4b,d possessing high specific activity have been prepared and tested as potential radiopharmaceuticals. The use of the 3-acetyl derivatives 2c,e and the replacement of iodine monochloride with sodium iodide and Chloramine-T in THF/phosphate buffer (pH 7.0) permitted us to synthesize no-carrier-added (17 alpha,20E)-21-(/sup 125/I)iodo-19-norpregna-1,3,5(10),20-tetraene-3,17-d iol (4b) and (17 alpha,20E)-21-(/sup 125/I)iodo-11 beta-methoxy-19-norpregna-1,3,5(10),20-tetraene-3,17-diol (4d) with 50% radiochemical yield and high purity. Although the specific activity represents only half of the theoretical value in some cases, this modified approach is a substantial improvement over the previously published method. Our preliminary distribution studies indicate that although both 4bmore » and 4d localize in the tissues known to have a large concentration of estrogen receptors, 4d accumulates in higher amounts in target tissues and provides a high target to nontarget ratio.« less
  • The possibility of the one-step synthesis of 17-spirooxetan-20-ones from 17..beta..-acetyl-17..cap alpha..-hydroxy steroids with the aid of iodosobenzene diacetate in a methanol solution of alkali was demonstrated.
