Effect of verapamil and OKY-046 on phorbol myristate acetate (PMA)-induced lung injury

In isolated dog lungs perfused at constant pressure, PMA (25-50 ..mu..g) was injected into the recirculating perfusate (vol = 550 ml), and its effect on vascular permeability, as determined by filtration coefficient (Kf) and isogravimetric capillary pressure (Pci), and on pulmonary vascular resistance (PVR) was observed. Group 1 lungs (n = 5) were perfused with plasma/dextran solution; Group 2 lungs (n = 6) were perfused with blood; Group 3 (n = 5) were blood-perfused and pretreated with 5 mg verapamil; and Group 4 (n = 3) were blood-perfused and pretreated with 100 mg OKY-046. Baseline (BL) values for Kf (g/min/cmH/sub 2/O/100g lung), Pci (cmH/sub 2/O), and PVR (cmH/sub 2/O/L/min/100g lung) are compared to those obtained one hour (1 Hr) after PMA (p < 0.05). The authors conclude that cellular components of blood are required for the permeability injury but not the increase in PVR, and the increase in permeability can be attenuated by a calcium-channel blocker or a thromboxane synthetase inhibitor.
