Enhancement of oxygen toxicity by diethyldithiocarbamate on Chinese hamster and mouse cells

A Cu/sup + +/-Zn/sup + +/ containing enzyme, superoxide dismutase (SOD) converts the toxic O/sub 2//sup -/ radical to molecular O/sub 2/. This radical may play a significant role in the effects of radiation and anti-cancer agents on cellular damage and lethality. Based on the assumption that inhibition of SOD activity should lead to enhanced cellular damage and lethality by radiation and certain anti-cancer agents (i.e., bleomycin and adriamycin) we exposed Chinese hamster and mouse cells to a Cu/sup + +/-chelator diethyldithiocarbamate (DDC) to reduce intracellular SOD activity. Treated cells consistently showed a lower level of survival. Irradiation of DDC-treated cells reduced even further the relative survival of these cells. Cytological determination of DDC-treated cells showed two abnormalities, i.e., (1) distortion and swelling of mitochondria and (2) myelin-like figure formation. The results suggest that it may be potentially beneficial in tumor therapy to combine some conventional cancer therapy modalities with an agent such as DDC in lowering the cells' defense against O/sub 2//sup -/ toxicity.
