Prolactin signal transduction: involvement of protein kinase C and CA/sup 2 +/

Phorbol esters (PE) in combination with calcium ionophore (A23187) mimic mitogen stimulation of proliferation in lymphocytes. This suggests that protein kinase C (PKC) activation and Ca/sup 2 +/ mobilization may be coupled to cellular proliferation. The mechanism by which prolactin (PRL) triggers a mitogenic response in target tissues is unclear. Therefore, a relationships of PKC activation and Ca/sup 2 +/ mobilization to ornithine decarboxylase (ODC) induction and /sup 3/H-thymidine (/sup 3/H-TdR) incorporation were studied in a rat node lymphoma cell line (Nb/sub 2/) dependent upon PRL for mitogenesis. PE enhanced PRL-stimulated ODC induction and /sup 3/H-TdR incorporation in Nb/sub 2/ cells. Addition of A23187 to cultures containing PE plus PRL increased ODC above PRL alone or PRL plus PE but attenuated proliferation compared to PRL plus PE. PE or PE plus A23187 increased /sup 3/H-TdR incorporation similar to low dose PRL, however this regimen was only 20-25% as effective as optimal PRL. PKC and calmodulin antagonists inhibited both PRL-stimulated ODC and proliferation. A cyclic AMP analogue also inhibited ODC induction and proliferation stimulated either by PRL or PE and A23187. These data strongly suggest that PKC activation and Ca/sup 2 +/ mobilization are coupled to PRL-stimulated ODC induction and proliferation in Nb/sub 2/ cells.
