THE INDEPENDENCE OF THE RADIOPROTECTIVE EFFECT OF AMINOETHYLISOTHIURONIUM. Br. HBr OF THE "OXYGEN EFFECT" (in Russian)

A relatively stable derivative of thiourea, 2-aminoethylisothiuronium (AET). Br. HBr, was used as a protective agent in mice irradiated with single, Co/sup 60/, gamma doses of liter, glass containers having a controlled oxygen atmosphere of 6.5, 8.2, 21 (air), 50, and 98.2% oxygen diluted wtth nitrogen. A concentration of 0.5 vol.% CO in air was also used. The percentage of survivals and length of survival were taken as criteria of the protective action of the AET compound. At a dose of 900 r the survival in air alone was zero, while the survival with 0.5% CO in air was 83.2%. The survival with a 10 mg injection of AET in air was 26.0%, and the survival with a 10 mg injection of AET in air having 0,5% CO was 93.5%. These results show that the protective effect of AET in combination with CO exceed the protective effect of CO alone. The survival in an 8.2% O/sub 2/ atmosphere was 48.4%, while with an 8.2% O/sub 2/ atmosphere plus a 10 mg injection of AET the survival was 78.1%. The controls irradiated in air at 900 r all died in this series of experiments. The protective effect of AET was greater at higher oxygen concentrations. Thus, the survival was-only 3.3% at 98.2% O/sub 2/, but it was 22.4% at 98.2% 0/sub 2/with the injection of 10 mg of AET into the mouse. Polarographic studies showed that the oxygen concentration in the tissues of the mice rose and fell with an increase or decrease of oxygen in the controlled atmosphere. It is concluded that the protective action of AET is independent of the oxygen effect. (TTT)
