THE EFFECTS OF AET (S,2-AMINOETHYLISOTHIURONIUMBROMIDE) ON THE INDUCTION OF DOMINANT LETHAL MUTATIONS BY X-RAY

One group of male mice (CBA x C3H and its reciprocal F/sub 1/ mature mice) was treated with AET 250 mg/kg 30 min beore local x-irradiation of 500 r over the testicular region. In another group this region was exposed without AET treatment. After irradiation each male was mated with two virgin females. The females, in which vaginal plugs were present, were taken away and substituted by other virgin females. The pregnant females were killed after 14 days' gestation and examined. The uteri were opened, and the number of living and dead embryos and corpora lutea was determined. Both the AET-treated and untreated males became sterile 4 weeks after irradiation and became fertile emryos/number of embryos implanted) and (nurnber of embryos implanted/number of corpora lutea) were compared in these 2 groups. lt was found that the ratios were not significantly different in the 2 groups, while the ratios were .very low compared with those of the control group. AET could not protect the induction of dominant lethal mutation in male mice. The reason for the ineffectiveness is considered to be the low concentration of AET in the reproductive cells at the time of irradiation. (auth)
