Morphologlc changes of MTK ascites tumor after x irradiation in vivo and in vitro were studied by the electron microscope. Immediately after in vivo irradiation, swelling of mitochondria, aberration of crista mitochondriales, and decrease of density of mitochondrial matrix were scen. They recovered, however, after several hours. The endoplasmic reticulum was imaged clearly just after irradiatlon of 200 r and was the most marked when 1000 r was given. Alter in vitro irradiation, changes similar to those observed in vlvo were seen, though very little recovery occurred. It was concluded that one of the mechanisms of the action of radiation on tumor cells consisted of direct injury to cell structares such as the destruction of mitochondria or endoplasmic reticulum.
