THE KINETICS OF STRONTIUM METABOLISM IN THE SKELETAL SYSTEM AND THE MECHANISM OF BINDING OF STRONTIUM IN BONE

Comparative studies were made of the kinetics of Sr and Ca uptake by bone in immature and adult rats fed these elements for various periods up to 120 days. Numerous tables are provided showing Sr and Ca contents of compact and porous regions of femur and tibia of male and female rats of various ages. Changes in exchangeable fractions of Sr and Ca were found to follow an exponential course and to be inversely related to each other, i.e., uptake of Sr by bone depends on Ca removal and Ca uptake on Sr removal. After various periods of Sr feeding, the Sr + Ca level in serum remained practically the same as the initial Ca level. Equilibrium constants for the Sr-Ca exchange reactions were calculated. The half-time values (in days) for Sr elimination from bone of young male and female rats fed Sr for 30 days were 28.2, 24.7; young rats fed Sr for 120 days 34.6, 30.1; and adult rats fed Sr for 120 days 204, 157. Thus Sr is eliminated from bone more slowly in grown than growing animals, whereas bone of growing animals shows a 2-3 times greater capacity for Sr incorporation. Spongiosa took up more Sr than compact bone, but elimination of Sr occurred at equal rates in both types of bone. The results indicate that Sr is taken up at the surface of bone mineral by an ion-exchange reaction. A portion of the stored Sr is firmly bound in the skeleton and unavailable for free exchange, apparently because of interference by organic bone matrix in the exchange process. The data provided no evidence that Sr replaces Ca in the crystalline lattice of bone. (H.H.D.)
