Radiogold (Au/sup 198/) seeds (0.8 x 2.5 mm) were implanted in the hypothalamus of adult guinea pigs and the brains were examined histologically 30 min to 102 days later. The seeds varied in activity from 3 to 10 mc and controls were implanted with inactive gold. Implantation of seeds produced only slight bleeding at early stages and minimum scar tissue formation at late stages. However, after 1 to 2 days the active seeds induced marked hyperemia and edema in surrounding brain tissue, which was followed by leukocytic infiltration. Necrotic changes could be noted with 40 hr and by 4 to 6 days extended up to 1 mm beyond the implantation site. Ganglion and glial cells (especially macroglia) showed degenerative changes, and these changes depended in intensity on the radiation dose, calculated to to vary beUeen 7,000 and 150,000 r. Edematous changes occurred in hypothalamus with 7,000 to 20,000 r and necrosis at doses above 30,000 r. At about the 14th day cyst formation began around the necrotic zone. Necrosis was considered to be a direct result of radiation on neural parenchyma, in spite of thc known relatively high radioresistance of the central nervous system. (H.H.D.)
