The mechanism of macromolecular protection was investigated in 0.02 mg/ ml concentrations of aqueous pepsin solutions (acidified with hydrochloric acid to pH 4.5 to 4.6) irradiated with Co/sup 60/ gamma rays. Damage was determined by proteolytic activity. Sodium nembutol, cystamine, sodium methabisulfite, AET, and BETA alanine were used as protective chemicals. Pepsin inactivation in the presence of cystamine is plotted as a function of various protector concentrations. A certain correlation was observed between the molecular dimensions of the protective materials and their blocking ability, the larger the molecule the smaller the concentration required to achieve a given degree of protection.
