ATTEMPTS TO DELINEATE INBORN ANEMIAS IN MICE. Progress Report, February 1, 1961-January 31, 1962
Results are reported from extensive investigations of the etiology of three different types of hereditary anemia in the house mouse. Studies of W- series macrocytic anemia include tests of potentialities and limitations of therapy through implantation of normal blood-forming tissue; detailed studies of contrasting reaction of normal and anemic mice to radiation damage; reaction of normal and anemic mice to different erythropoietic stimuli and to plethorization; and search for metabolic error in hematopoiesis through isotopic-labeling studies of the kinetics of heme synthesis in normal and genetically anemic mice. Studies of Sl-series macrocytic anemia include characterization of effects of a new viable allele (Sl/sup d/); attempts at therapy through implantation; and studies of reactions of viable anemic mice to erythropoietic stimuli and to x irradiation. Similarities and differences between results with the two genetically distinct macrocytic anemias may provide valuable clues as to the gene-actions involved. Studies of the transitory siderocytic anemia of ff mice, based on a new homozygous inbred stock (F L/Re), include detailed characterization of tide anemia and its effects on growth in fetuses and newborn mice; tests of effects of single-gene substitutions (Ww, Ff, and FF) on this syndrome; and search for evidence of metabolic error in FL/Re mice. The previous finding of effects of single-gene substitutions on acute radiosensitivity is being followed up by a more extensive study of the effects of single doses of a wide range of deleterious genes affecting hematopoietic tissue. Studies of the nature and mechanism of action of a specific radiation protection factor are continuing. Additional areas of investigation include characterization of new hereditary anemias as they arise and preparation of controlled genetic stocks for further study of such new material. (auth)
