STUDIES ON THE ELECTRONIC PARAMAGNETIC RESONANCE OF DEOXYRIBONUCLEIC ACID EXTRACTED FROM BACTERIOPHAGE T2

Measurements were made on deoxyribonucleic acid (DNA) from calf thymus and from Escherichia coli T2 phage, before and after irradiation of the DNA with doses up to 5 x l0/sup 6/ r. The electron paramagnetic resonance spectra of native and irradiated T2 DNA showed pronounced differences, and irradiated T2 and calf DNA were also different. Native calf DNA showed resonance line centered at g = 2.00 and at 1800 gauss and above, whereas native T2 DNA showed no resonance lines. Irradiated T2 DNA, in comparison with irradiated calf DNA, showed a Hyperfine spectral structure formed by 3 equidistant lines of equal intensity. The signal appeared to be superimposed on a larger line lacking hyperfine structure, centerial about 30 gauss. It is suggested that this difference may be attributed to the presence of 5hydroxymethylcytosine in T2 DNA in contrast to cytosine in calf DNA and that the former purine may be the site of x-ray inactivation of T2 phage DNA.
