Radiosensitization of a Mammalian Cell Line with 5-Bromodeoxyuridine
BS>Some radiobiological properties of a mammalian cell line after exposure to the halogenated nucleoside 5-bromodeoxyuridine (BUDR) are described. Incorporation of BUDR into the deoxyribonucleic acid of mammalian cells in culture to the extent of 1% was measured after 3 to 4 days of in cubation with 6 mu g of the analog per milliliter. An in creased x-ray sensitivity of about 2 and an increase in extrapolation number of survival curves were observed after exposure to the above-mentioned concentration of BUDR. In contrast to this, an enhanced sensitivity slightly greater than 1 and a decrease in extrapolation number were found after thermal neutron irradiation and identical treatment with BUDR. Theoretical calculations on x-ray energy absorbed by BUDR-exposed cells show that a negligible increase in energy is absorbed per cell with incorporation of 1% BUDR, as compared to unexposed cells. For analogexposed cells, survival was found to increase when a single dose of x rays was administered 4 to 12 hours after cell plating. For unexposed cells, survival was found to be constant during that same period.
