X-irradiation and Diphosphopyridine Nucleotide (DPN) Levels in Ascites-tumour Cells and Liver Tissue of Mice
Mice with Ehrlich's ascites tumor received total-body x irradiation with 900 to 5000 r. Concentrationa of oxidized DPN were analyzed enzymatically with yeast alcohol dehydrogenase in liver and tumor-cell homogenates 2 hr and 12 hr after irradiation. After 5000 r, tumor cells and liver exhibited but little decrease of DPN, 5% and 7% respectively at 2 hr, and 12% at 12 hr. When animals were irradiated before or after intramuscular administration of nicotinamide (170 mg/kg), significant alterations of steady-state concentrations of DPN were observed. After irradiation, average decreases of DPN in tumor cells and liver were 22% and 27% respectively (2 hr) and 16% and 17% (12 hr). It follows that x irradiation within the stated dose-range moderately inhibits DPN-synthesis or activates DFNbreakdowm. The effect is compensated at normal cellular oncentrations of DPN, but is demonstrated if DPNynthesis is activated by administration of nicotinamide.
