Electro-shock Seizure Responses to Reserpine and Iproniazid after X-irradiation in Rats

Reserpine (2 mg/kg) and iproniazid (100 mg/kg) were administered to adult rats after single doses of 500 r whole-body x irradiation, Threshold for minimal electroshock seizures and pattern of maximal electro-shock seizures were studied under experimental conditions of drug injection and irradiation. X irradiation lowers threshold for minimal convulsions which suggests an increased brain excitability; it also shortens maximal tonic-- clonic convulsions, which in turn can be interpreted as a more rapid than normal onset of neutral fatigue. Reserpine has a synergistic effect on the post-irradiation fall in threshold while counteracting the shortening of the maximal seizure. Iproniazid has a less marked synergistic effect on the fall in threshold after irradiation and produces no significant change in the pattern of maximal convulsions. These results were interpreted in terms of a direct action of irradiation, reserpine, and iproniazid on brain function involving mechanisms related to brain serotonin. (auth)
