The distribution of calcium in the epiphyseal cartilage of the rat tibia measured with the electron probe X-ray microanalyzer

A description is given of the electron probe x-ray microanalyzer used to determine the Ca distribution in the epiphyseal plate of the rat tibia. Vacuum- coating of the speciMen with a thin layer of Al was added to the usual techniques for preparing thin section of biological specimens. Ca content was found to change from about 30 to 0% from the completely calcified cartilage edge toward the maturation zone of the epiphyseal plate. Variation in the concentration gradient may be related to the nonuniform distribution of hydroxyapattte crystals in the matrix collagen. (H.H.D.)
