Some Effects of Parathyroid Hormone on the Metabolism of Sr 85 and Ca 45 by Embryonic Chick Bone Cultured in Vitro

Embryonic chick bone was cultured in vitro in a synthetic medium (N.C.T.C.-109) to test the effect of parathyroid extract (PTE) on the metabolism of Ca45 and Sr85. When contrasted with the control bones, PTE at levels of lation of Sr85 and Ca45 in the bone and increased Observed Ratio (bone/medium). The low levels of PTE increased, while the very high levels of PTE decreased, both the growth and mass of the bones. The short term uptake of Sr85 and Ca45 was unaffected by pretreatment of the bone with PTE except at a level of 10 units/ml. PTE at 0.1 and 1.0 unit/ml did not change the proportion of labile to nonlabile Sr85 and Ca45 in the bones. The nonlabile fraction could be readily mobilized by the addition of PTE into the medium, with 0.1 and 1.0 unit/ml having an apparently equal effect. Sustained PTE treatment of the bones increased the mobility of the nonlabile fraction over that observed after a relatively short exposure to the hormone. PTE acted to reduce the isotope content of growing bone by increasing the movement of mineral from bone to medium, and thus reduced the size of the mineral compartment.
